Characterizing Interactions of Tolfenpyrad with Antibiotics Against Francisella novicida: Synergy, Antagonism,
and Mechanistic Insights

Research Description:

Francisella tularensis is a highly virulent Gram-negative bacterial pathogen that can be transmitted via aerosols and arthropod bites. As one of the most infectious pathogens known, as few as 10 viable bacteria can cause a deadly disease. This highlights both its potential use as a bioweapon and the urgency to develop new strategies to safeguard against antimicrobial resistance. Thus, it is essential to innovate novel and improve existing treatments against Francisella. We recently identified a pesticide, tolfenpyrad, that potently blocks Francisella growth; however, the mechanism that tolfenpyrad employs to disrupt Francisella growth is unknown. This research aims to shed light on tolfenpyrad's mechanism of action.

Project Responsibilities:

Conduct plate-based assays with combinations of tolfenpyrad and characterized antibiotics to assess synergism and antagonism.
Utilize flow cytometry to assess fluorescence of tetracycline when in combination with tolfenpyrad.
Perform basic lab duties, such as producing agar plates and proper biohazardous waste removal.
